It depends. Do you have a reg? What's the psi it's running on? But, I think you should have done some research before you made such a big investment.

The rubber thing is a thread protecter, just unscrew it like a bottle cap.

Then insert in the same place the C02 was, turn clockwise, cock gun, pull trigger. Once empty re-fill and repeat.

I take it your tank is probally steel, or it might be fiber wrapped, but otherwise, a C02 tank and a Nitrogen or Compressed Air Tank are different, but the way you connect and remove them to and from the gun are the exact same way. The only difference between a C02 Tank and a Nitro Tank is that a nitro tank has a built on regulator that lets the air out at a set pressure that can be adjusted or it comes preset. A Nitro tank is built to accomadate Nitrogen or Compressed air. It also comes with a fill nipple so you can fill your tank without removing from your marker (as long as you have an on/off). It also comes with a guage because Nitrogen is not measured in ounces as C02 so when filling your tank it is not as much as a "guess how much to put in and weigh" filling procedure as with C02. You look at the guage and are able to see when it is full. A C02 tank is simply a tank that has no regulator and out puts at whatever the gas does. It is only meant for C02 purposes. That is why there is such a huge prices difference.

I know this is lengthy and is really uneccisary but I wanted to clear everything up for you(plus I wanted to sound really smart). Hope it helps. If anyone finds anything wrong with my info please post any correction.
